What size base should they be on?

40mm round bases would do the trick.

and therefore they would take up 4 squares?

A big guys uses only one square on the pitch. So say the rules.

But it happens that they use more squares in real life, cause they have a big base.

Make a base to fit.

a 40mm round base fits pretty well into a 29mm bloodbowl square. the few mm it reaches into neighboring squares can be tolerated because most other players only have 25mm bases.

Since my BB Big Guys are a Warhammer Rat Ogre from my Skaven Army and a (unconverted) Mordheim ogre, they use 40mm square bases which are even larger.
This also works good enough, they are taking up a bit more space than a Bloodbowl field square, but there is enough space around them so that you can easily place all other players.

The bigger problem is if you have very large models and they fall over, things might get a bit crowded. If you experience problems with that, you might want to start using a marker for your prone player.

In addition to what Tim said, we use markers when the Biggies are down. Especially with his two cows and two Trolls, the pitch sometimes gets a bit crowded

It never seems to cause any problems unless You have an ogre team trying to form a cage. That really really sucks.
